As different tokens have proliferated over the years, the viable options have increased as well. While there may have close to an optimal set up back in the old (wooden) days, players now have many many choices as to their setup and play-style.
A good starting point is with your own tokens- If you're a purpled-out UR player, you'll have a different setup then someone aiming for Reds. Both work. Think about how you've seen others play in TD, the encounters from prior years, and what you like out of your PCs in other fantasy games.
An example: In TD, some fighters go for a reciprocal damage approach- lots of Amulet of the Thorns kinda items. Other people run with a high AC, and plan not to be hit. Some max out STR, to take out the monster first. Others try to balance as much as possible, not knowing what the dungeon will throw their way. All could work.
Me, I run a ranged Rogue. Great dex, horrible strength, and forgo the Sneak Attack. I'm greedy, so I'd rather take the risk by using a Ring of Riches instead of a something else. I've sacrificed AC for increased Saves. Maybe not it is not optimal, but it is a style I've fined-tuned, and really like.
